Etanol označován také jako ethylalkohol, je druhý nejnižší alkohol. Je to látka s účinky na
lidskou psychiku. Tato diplomová práce je ve své teoretické části soustředěna na chemickou
strukturu, biologickou produkci a biochemii mikrobiální produkce etanolu. Podstatná část je
věnována praktickému významu etanolu pro lidstvo, průmyslové výrobě etanolu a především
působení etanolu na mikroorganismy. Závěr teoretické části je pak věnován kvasince
Saccharomyces cerevisiae a její roli v produkci etanolu.
Praktická část diplomové práce je zaměřena na izolaci mikroorganismů z volné přírody a dále
pak na hrubé stanovení koncentrace etanolu, při které jsou izoláty mikroorganismů ještě
schopny růst. Tato koncentrace pak byla použita v nahromaďovacích kulturách, které sloužily
k izolaci mikroorganismů tolerujících etanol. Celkem bylo tímto způsobem získáno 8
takových izolátů kvasinek. Následně byla provedena jejich identifikace a charakterizace.
Z výsledků praktické části vyplývá, že kvasinky nacházející se ve volné přírodě dokáží do
určité míry tolerovat přítomnost etanolu. Tato schopnost, ale nesouvisí se schopností
produkovat etanol anaerobní fermentací.

Ethanol, also known as ethyl alcohol, is the second lowest alcohol in the world. It is a
psychoactive substance. The theoretical part of this diploma thesis is focused on the chemical
structure, biological production and biochemistry of microbial ethanol production. A
substantial part is devoted to the practical importance of ethanol for humanity, industrial
production of ethanol and especially the effect of ethanol on microorganisms. The final
section of the theoretical part is focused on the yeast Saccharomyces cerevisiae and its role in
ethanol production.
The practical part of the diploma thesis is focused on the isolation of microorganisms from
nature and on rough determination of the concentration of ethanol at which the isolates were
still able to grow. This concentration was used in enrichment cultures established in order to
isolate the micrrorganisms that tolerate ethanol. This way, a total of 8 yeast isolates were
obtained. Subsequently, the identification and characterization of the obtained isolates were
performed.
The results of the practical part show that yeasts found in nature can tolerate the presence of
ethanol to some extent. This ability, however, is not related to the ability to produce ethanol
by anaerobic fermentation.

2) Vzorky materiálu, který bude sloužit jako zdroj mikroorganismů pro experimenty, budou získány z přírody. Strategie volby prostředí, z nichž budou tyto vzorky získány, bude konzultována s vedoucím práce.
3) Zvolené laboratorní postupy budou konzultovány s vedoucím práce.

1) Stanovení inhibiční koncentrace etanolu u členů přirozených mikrobiálních společenstev
