Extracelulární vezikuly jsou malé membránové částice (30 nm - 5 ?m) uvolňované třemi základními doménami života - bakteriemi, eukaryoty i archei. Ukazuje se, že extracelulární váčky hrají významnou roli při mezibuněčné komunikaci, a to jak v rámci jednotlivých říší, tak i mezi nimi. Výzkum se zabývá převážně extracelulárními váčky sekretovanými různými typy živočišných buněk, včetně buněk člověka. Váčky regulují mezibuněčnou komunikaci prostřednictvím dopravy bioaktivních molekul karga a mohou se tak podílet na změně genové exprese, regulace buněčných dějů a fyziologických procesů recipienta. Ukázalo se, že extracelulární vezikuly mohou sloužit k cílené dopravě léčiv i jako biomarkery různých onemocnění, jako je např. rakovina.
Různé typy extracelulárních vezikulů byly identifikovány v savčích buňkách, kde hrají důležité role. Dlouhou dobu se spekulovalo o výskytu extracelulárních váčků u rostlin kvůli přítomnosti buněčné stěny na povrchu rostlinných buněk, která by mohla bránit uvolňování a přijímání rostlinných váčků. Ukázalo se, že rostliny skutečně produkují extracelulární vezikuly mající různé funkce, jako je sekrece proteinů, transport bioaktivních molekul nebo obrana rostlin proti patogenům. Zdá se, že se rostlinné extracelulární váčky mohou podílet na dopravě léčiv stejně jako váčky živočišné. Zájem o poznání jejich vlastností, funkcí a možného využití v posledních letech roste.

Extracellular vesicles are small membrane particles (30 nm - 5 ?m) released by three main domains of life - Bacteria, Eukaryotes and Archaea. Extracellular vesicles have been shown to play an important role in intracellular comunication, both within and between kingdoms. The research focuses mainly on extracellular vesicles secreted by various types of animal cells, including human cells. These vesicles regulate intercellular comunication through the transport of bioactive cargo molecules. Thus, they can be involved in altering gene expression, regulation of cellular and the physiological processes of the recipient. It has been shown that extracellular vesicles can also be used for targeted drug delivery and as a biomarkers of various diseases, such as cancer.
Different types of extracellular vesicles have been identified in mammalian cells where they play an important roles. For a long time there had been speculations about the presence of extracellular vesicles in plants due to the presence of a cell wall that could inhibit the release and uptake of plant vesicles. It has been already shown that plants do produce extracellular vesicles having various functions such as protein secretion, transport of bioactive molecules or plant defense against pathogens. It appears that plant extracellular vesicles may be involved in drug delivery as well as animal vesicles. The interest about their properties, functions and their possible use has been growing in recent years.

1. V rámci teoretického úvodu vytvořit rešerši shrnující dosavadní znalosti o rostlinných extracelulárních váčcích (ECV) a srovnat je se znalostmi o živočišných ECV.
2. Optimalizovat vybraný protokol na izolaci exosomů u N. tabacum.
3. Navrhnout možné metody analýzy charakterizace (velikosti a složení ECV u rostlin) a testovat vybranou metodu charakterizace ECV na vyizolovaných vzorcích.
Zásady pro vypracování
1. V rámci teoretického úvodu vytvořit rešerši shrnující dosavadní znalosti o rostlinných extracelulárních váčcích (ECV) a srovnat je se znalostmi o živočišných ECV.
2. Optimalizovat vybraný protokol na izolaci exosomů u N. tabacum.
3. Navrhnout možné metody analýzy charakterizace (velikosti a složení ECV u rostlin) a testovat vybranou metodu charakterizace ECV na vyizolovaných vzorcích.
